Mossdog is then targeted by opponent's spell/ability.

Does Mossdog die with a +1/+1 counter on it or does spinal graft's destroy effect kill mossdog before a counter can be placed?

Depends who controls Spinal Graft and whose turn it is.

If the same player controls both Mossdog and Spinal Graft, that player can choose which order they resolve in, choosing whether or not Mossdog gets a counter before dying.

If a different player controls them, then it depends on whose turn it is. If it's Mossdog's controller's turn, then Mossdog's ability goes on the stack first, then Spinal Graft. Graft resolves first and kills Mossdog before the counter is put on. If it's the other player's turn, it's the opposite.
