    Is it ok to regenerate a creature that hasn't taken lethal dmg?

    Sure. You can activate a regeneration ability whenever you want (well, whenever you can play an activated ability at least). It just won't take effect until the creature would actually be destroyed.

    Matter of fact, if you wait until the creature has already taken lethal damage, it's too late.

    The regeneration ability says "The next time ~ would be destroyed this turn...", so no.

    The shield only lasts for one turn - if it hasn't been used by the end of the turn, it disappears.
    Fluttershy is best pony.
