Depends what exactly you mean. If you're targeted by something like Blightning, then it'll do as much as possible when it resolves - you can't discard anything, so you'll just take 3 damage. If you have to pay a cost that involves discarding, such as for Wilf Mongrel or Sonic Seizure, then you can't pay that cost and therefore can't activate the ability/cast the spell.
Fluttershy is best pony.

Since they can't choose to discard, they have to lose 5 life.
